Transgenic Mice

   

Transgenic mice are constructed by injecting cloned DNA into fertilized eggs; those eggs that survive are then implanted in foster females to develop to term. TCDM will ear-punch the mouse pups and collect tail and/or ear punch biopsies for genotyping by Southern blotting and/or PCR. For transient transgenics (examining DNA integration and expression in transgenic embryos), the pregnant females are dissected on the desired date.

The main factor affecting the production efficiency of transgenic mice is the quality of DNA preparation. TCDM will provide you with protocol for high quality DNA purification.

Potential founders will be screened by investigators for the presence of the transgene by PCR or southern blot genotyping assays. It is highly recommended to establish a robust screening method capable to detect a transgene when it is spiked into tail DNA at a single copy concentration BEFORE submitting DNA for microinjection.

If the studies on tail biopsy DNA (with adequate controls) fail to demonstrate the presence of at least three transgenic founder animals, we will reinject the construct second time at no additional cost to the investigator. We do not guarantee expression of the transgene. A founder is defined as any mouse with either the full or partial DNA construct.
